The budget supermarket chain first announced plans to open a store in the town three years ago.
The planning application has been hindered by environmental concerns due to its proximity to the River Misbourne.
An Aldi spokesperson said: “Aldi knows its customers are currently travelling out of the Amersham area to visit its stores. It has wanted to open a local store in Amersham for some time and is delighted to have identified the former Jaguar Dealership at 44 London Road West as a location.
“This significant investment would create at least 40 new jobs, which Aldi would seek to recruit locally. The site is a disused brownfield site and will be landscaped to complement the local area. There will be 101 parking bays, parking for Blue Badge holders and parents and children. There will also be cycle parking and four electric vehicle charging points.”
The fate of the proposed Aldi will now be decided at a Buckinghamshire Council meeting on April 30 after the initial Planning Committee meeting scheduled for April 2 was postponed.
